Ebola virus protein VP40 induces accessory cell-mediated activation of human natural killer (NK) cells

Abstract Natural killer (NK) cells have been implicated in Ebola virus disease (EVD) pathogenesis as well protection against viruses. A clear understanding of their role during EVD is needed for better exploiting functionalities designing effective countermeasures Previously, we reported that protein VP40 induces IL-12 and IL-18 dependent activation human NK involves accessory cells. Here, extended our studies to define the identities molecular mechanism(s) underlying triggering cell activation. Purified CD14 +myeloid cells, CD3 +T or CD19 +B isolated from normal peripheral blood mononuclear (PBMCs), were co-cultured with purified CD56 +NK cells; cultures activated expression IFN-γ, granzyme B, perforin, surface CD107a was measured. small, but significant, increase observed when alone incubated VP40.
